HygromycinBinhibitsproteinsynthesisbystrengtheningtheinteractionoftRNAbindingintheribosomalA-site.HygromycinBalsopreventsmRNAandtRNAtranslocationbyanunknownmechanism.TheseareuniquemechanismsforanaminoglycosideantibioticandtheydifferfromthemodeofactionofNeomycin,Gentamicin,andG418. MechanismofResistance HygromycinBresistanceisconferredbythehphgeneisolatedfromStreptomyceshygroscopicus,a1467bpfragmentwhichencodesHygromycinBphosphotransferase(HPh).CelllinessuccessfullytransfectedwiththehphgeneproducehygromycinBphosphotransferaseandconverthygromycinBto7”-O-phosphoryl-hygromycinBbyphosphorylatingthe4-hydroxylgrouponthecyclitolringofhygromycinB.7”-O-phosphoryl-hygromycinBlacksantibioticactivityanddoesnotinteractwithprokaryoticoreukaryoticribosomes.
