MechanismofAction
β-lactamsinterferewithPBP(penicillinbindingprotein)activityinvolvedinthefinalphaseofpeptidoglycansynthesis.PBP’sareenzymeswhichcatalyzeapentaglycinecrosslinkbetweenalanineandlysineresiduesprovidingadditionalstrengthtothecellwall.Withoutapentaglycinecrosslink,theintegrityofthecellwallisseverelycompromisedandultimatelyleadstocelllysisanddeath.Resistancetoβ-lactamsiscommonlyduetocellscontainingplasmidencodedβ-lactamases.Likemanycarbapenems,meropenemishighlyresistanttothedegradativeeffectsofβ-lactamases.
