Streptomycinsulfateisoftenusedtogetherwithpenicillinandotheragentstoinhibitbacterialcontaminationincellcultureapplications.ItisrecommendedforuseinmolecularBIOLOGyapplicationsat25-50μg/mL,incellcultureapplicationsat100mg/Landinembryocultureat50mg/L.

MechanismofAction
Streptomycinisaproteinsynthesisinhibitor.Itbindstothesmall16SrRNAofthe30Ssubunitofthebacterialribosome,interferingwiththebindingofformyl-methionyl-tRNAtothe30Ssubunit.Thisleadstocodonmisreading,eventualinhibitionofproteinsynthesisandultimatelydeathofmicrobialcellsthroughmechanismsthatarestillnotunderstood.Speculationonthismechanismindicatesthatthebindingofthemoleculetothe30Ssubunitinterfereswith50SsubunitassociationwiththemRNAstrand.Thisresultsinanunstableribosomal-mRNAcomplex,leadingtoaframeshiftmutationanddefectiveproteinsynthesis;leadingtocelldeath.
