Theaminocyclitolantibioticspectinomycin,oftenconsideredalongsidetheaminoglycosides,bindsinreversIBLefashion(hencethebacteriostaticactivity)tothe16SrRNAoftheribosomal30Ssubunit.ThereitinterruptsthetranslocationeventthatoccursasthenextcodonofmRNAisalignedwiththeAsiteinreADInessfortheincomingaminoacyl-tRNA.Structuralstudiesrevealthattheantibioticbindstoanareaofthe30Ssubunitknownastheheadregionwhichneedstomoveduringtranslocation.Bindingoftherigidspectinomycinmoleculeappearstopreventthemovementrequiredfortranslocation.
